I'm not sure either, but maybe it's asking us to pick out 12 samples (1 year of data), for each year.

In question 5b, what do the question mean by asking us to remove the annual cycle? Is the question asking us to remove the current year sample or last 12 sample from the data?

I'm not sure either, but maybe it's asking us to pick out 12 samples (1 year of data), for each year. When you have that data, you can then average each year and tell if your temperature is changing per year.
